Standards
Most CD instruments require calibration with a CD standard for intensity. An Olis instrument does not require this, although it is comforting to run a known standard to occasionally confirm that the instrument is functioning properly. The most common standard used is (+)-10-camphorsulfonic acid (CSA). At a concentration of 1 mg/mL and a 1 mm pathlength, two peaks are observed. A positive peak at 290.5 nm exhibits an intensity of 31 mdeg (2.36 mA) and a negative intensity of 62 mdeg (5.0 mA) at 192.5 nm. An example of this spectrum is shown in Figure 6.
